Micropropagation for in vitro Culture of Saintpaulia at Controlled pH and Various Sucrose Levels



Abstract
Tissue culture of Saintpaulia icnantha Wendl. cultivar `Blue Boy¢¥ and `California¢¥ for a possible extensive propagation was attempted by applying different sucrose levels, sterilizations, plant materials, or pH levels for cultural medium. The experimental results obtained are shown as follows;
1. Application of 10% Clorax with 1% Tween-20 for surface sterilization showed the least microbial contamination of in vitro tissues.
2. Tissues taken originally from leaves grew much better than these from petioles.
3. Cultural medium with 6% sucrose gave the best results in tissue growth, while less so with 3% and least so with 1% sucrose. 6%-sucrose media w-as observed to show more root. differentiation than the other two sucrose levels.
4. pH 5.8 of cultural medium was found best suited for an improved growth of tissues. and a bit lower pH level seemed also preferable for the culture. pH toward basic levels was unfzvorable and pH 6.9 showed almost no onset of organ differentiation and produced only a brownish calli.
Regardless of pH adjustment prior to autoclaving, the media tended to stand in the range of pH 5.8, and this is regarded due largely to the buffering property of agar in the media.
